چکیده

The crystal structure of uridylyl 3’,5’-adenosine phosphate (UpA) has been determined and retied to an R value of O-062. There are two UpA molecules per asymmetric unit in 8 monoclinic unit cell with dimensions a = 16+91,6 = 12.37, c = 11-26 d and fi = 96.0'. The conformation of the phosphodiester linkage is different for the two molecules; one UpA suggests a model for a sharp turn in a single-stranded nucleic acid structure and the other for a helical structure. All the nuoleoside units have similar conformations. There is no complementary base pairing between the adenines and uracils but rather layers of base-paired adenines and base-paired uracils separated by 3.4 A. The predominant form of base stacking involves the ribosyl oxygen atom and the bases.

Structural basis for the fast self-cleavage reaction catalyzed by the twister ribozyme.

Twister is a recently discovered RNA motif that is estimated to have one of the fastest known catalytic rates of any naturally occurring small self-cleaving ribozyme. We determined the 4.1-Å resolution crystal structure of a twister sequence from an organism that has not been cultured in isolation, and it shows an ordered scissile phosphate and nucleotide 5' to the cleavage site. A second cryst...

Elements of Nucleotide Specificity in the Trypanosoma brucei Mitochondrial RNA Editing Enzyme RET2

The causative agent of African sleeping sickness, Trypanosoma brucei , undergoes an unusual mitochondrial RNA editing process that is essential for its survival. RNA editing terminal uridylyl transferase 2 of T. brucei (TbRET2) is an indispensable component of the editosome machinery that performs this editing. TbRET2 is required to maintain the vitality of both the insect and bloodstream fo...

Backbone conformations in secondary and tertiary structural units of nucleic acids. Constraint in the phosphodiester conformation.

The possible backbone phosphodiester conformations in a dinucleoside monophosphate and a dinucleoside triphosphate have been investigated by semiempirical energy calculations. Conformational energies have been computed as a function of the rotations omega' and omega about the internucleotide P-O(3') and P-O(5') linkages, with the nucleotide residues themselves assumed to be in one of the prefer...
